  We have an exciting opportunity for a Postdoctoral Researcher in Professor Adrian Harris’s laboratory at the Weatherall Institute for Molecular Medicine, Department of Oncology, Headington Oxford.

  The development of antiangiogenic therapy has really depended on work in xenografts and transgenic models. Little has been established about the molecular biology and gene expression profiles, heterogeneity and potential genetic changes in tumour vasculature by any modern technology. We hypothesise that there is great heterogeneity in the tumour endothelial population, that it is adaptive and may have a genetic basis.

  We will investigate this using state of the art methodology for endothelial separation and single cell sequencing at DNA level and RNA sequencing. We profile up to 1000 endothelial cells using the Chromium 10 apparatus, which can allow very high throughput sampling of up to 10,000 cells, 70,000 reads each in 24 hours, which will revolutionise our approach to endothelial biology from human cancer.

  Based on these findings we will investigate models for resistance mechanisms and roles of metastasis of newly discovered signalling pathways. There will also be links to clinical samples before and after antiangiogenic therapy to investigate mechanisms of resistance. The cell biology of these new pathways will be investigated using appropriate endothelial cells, either from primary tumours or from mice and we may need to proceed to make appropriate genetic models in the longer run. The expression of these genes will be assessed on the relevant clinical specimens and animal model systems by immunochemistry or RNAscope FISH.

  The postholder will be a postdoctoral level scientist holding or be nearing completion of, a PhD/DPhil in a relevant area. Expertise in cell and molecular biology will be a necessity, along with evidence of previous publications. A Personal Animal Licence would be advantageous for this role.

  This is a full-time post and is available until 30 September 2020 in the first instance.
